IN PHARBITIDIS SEMEN BY HPLC METHOD

Summary

High-performance liquid chromatography (HPLC) was employed to quantify chlorogenic acid (CGA) and isochlorogenic acid A (ICGA) in Pharbitidis Semen, owing to its high sensitivity and suitability for quantitative analysis. An Agilent Eclipse Plus C18 column (250 × 4.6 mm, 5 µm) and a DAD detector at 327 nm were employed for separation and detection. The mobile phase consisted of 0.1% formic acid in water and acetonitrile, with a flow rate of 1.0 mL/min and a 5 µL injection volume. The method demonstrated linearity over the ranges of 4.6 – 557.62 μg/mL for CGA and 4.3 – 557.62 μg/mL for ICGA. Validation parameters, including accuracy, system suitability, linearity, repeatability, precision, limit of detection (LOD), and limit of quantification (LOQ), met the AOAC requirements according to ICH guidelines. The findings are useful for quality control of Pharbitidis Semen in Vietnam.
