HPLC METHOD FOR THE SIMULTANEOUS DETERMINATION OF THREE LIGNANS OF SCHISANDRA SPHENANTHERA COLLECTED IN NGOC LINH MOUNTAIN, KON TUM PROVINCE, VIETNAM

Summary
HPLC Method for the Simultaneous Determination of three Lignans of Schisandra sphenanthera Collected in Ngoc Linh Mountain, Kon Tum Province, Vietnam
A simple and sensitive high performance liquid chromatography (HPLC-DAD) method for simultaneous quantification of schisandrin, gomisin B, schisandrin A in the ripe fruit of Schisandra sphenanthera was developed and validated. The chromatographic separations were obtained by an Agilent C18 column (250 x 4.6 mm i.d., 5 µm particle size) using gradient elution with acetonitrile and water, at a flow rate of 0.8 mL/min and detection wavelength at 254 nm. All calibration curves displayed a good linear relationship (R2=0.999). The validation results displayed good accuracy, repeatability, linearity, and selectivity according to the ICH guideline. The established method was applied to analyze 6 samples collected Ngoc Linh mountain (Kon Tum, Vietnam). The contents of schisandrin, gomisin B, schisandrin A in Schisandra sphenanthera samples collected in Ngoc Linh mountain were 0.197-0.246%, 0.207-0.275%, and 0.107- 0.132%, respectively. The obtained results provided useful information for the quality testing of the fruit of Schisandra sphenanthera in Vietnam.
