Summary
Expression of α-Synuclein Protein of Parkinson Disease on Drosophila Model and
Effect of Bacopa monnieri (L.) Wettst. on the Model
α-Synuclein is a presynaptic neuronal protein that is linked genetically and neuropathologically to Parkinson's disease (PD). α-Synuclein has been identified as the major component of Lewy bodies and Lewy neurites, the characteristic proteinaceous deposits that are the hallmarks of PD. The abnormal accumulations of fibrillar α-synuclein in Lewy bodies and the mutations in the gene for α-synuclein in familial forms of PD have led to the belief that this protein has a central role in a group of neurodegenerative diseases known as the synucleinopathies.
The fruit fly Drosophila melanogaster is a versatile model organism that has been used in biomedical research for over a century to study a broad range of phenomena. There are many technical advantages of using Drosophila over vertebrate models; they are easy and inexpensive to culture in laboratory conditions, have a much shorter life cycle, they produce large numbers of externally laid embryos and they can be genetically modified in numerous ways. Research using Drosophila has made key advances in our understanding of regenerative biology and will no doubt contribute to the future of regenerative medicine in many different ways.
In this study, we express α-synuclein protein in Drosophila model and investigated the effects of Bacopa monnieri (L.) Wettst. on this model. The adult flies carrying α-synuclein show rough eyes phenotype under gmr-gal4 driver. So far, expression of the protein was confirmed by immunostaining with anti α-synuclein antibody. The rough eyes phenotype induced by expression of α-synuclein were reduced by feeding Bacopa monnieri (L.) Wettst. of 0.1 mg/l compare to those without feeding Bacopa monnieri (L.) Wettst.
