Determination of Some Active Components in Fructus gardeniae by Thin Layer Chromatography - Scanning Method fof Standardization
Thin layer chromatography- fingerprint (TLC-FP) analysis of iridoids and crocin pigment and quantitative determination of geniposide, the major iridoid component on fructus gardeniae were performed. For TLC-FP identification of iridoids, the solvent system of N.butanol : acid acetic : nước ( 7 : 2 :1 ) and anisaldehyde- sulphuric acid reagent were used. For TLC-FP identification of crocin pigment, the solvent system of Ethylacetat: isopropanol: nước ( 6,5 : 2,5 : 1) was used; the detection is observed in day ligth and under UV 254 fluoresent. TLC plate silicagel 60 Merck was used for the identification . For quantitative analysis, the solvent system is the same one for the separation of crocin pigment, HPTLC plate silicagel 60 Merck was used and scanning was performed at l 235nm by Camag TLC scanner3. The calibration plot is linear in the range of 0,4 - 2,4 mg ( r : 0,99781). The repeatability and the average recovery of the method is 1,451% (RSD) and 103,0%, respectively. The methods take part in research standardization and quality control of Fructus Gardeniae.
