Development Method for Quantitative Determination of the Major Flavonoid 2′,4′-Dihydroxy-6′-Methoxy-3′,5′-Dimethylchalcone in the Buds and Leaves of Cleistocalyx operculatus by HPLC
Cleistocalyx operculatus have been used in Vietnamese and Chinese traditional medicine for a long. Previous studies indicated that the flavonoid 2′,4′-dihydroxy-6′-methoxy-3′,5′-dimethylchalcone is the major compounds in the buds and leaves of this plant. Therefore, this study aims with the quantitative determination of this compound in these materials collected in the north of Vietnam. An accurate, simple and sensitive method using HPLC with UV detection was developed for the analytical quantification of 2′,4′-dihydroxy-6′-methoxy-3′,5′-dimethylchalcone in buds and leaves of C. operculatus. Assay was conducted using a Bondapak C18 column (3.9 × 300 mm, 5 µm) under isocratic elution with NaH2PO4 0.05M/water – methanol (20:80) at flow rate of 0.7 ml/min and detected using photodiode array at 341 nm. The method showed good linearity in the range 5.5-110 µg/ml with the correlation coefficient r2 = 0.9982, as well as good recovery and accuracy. The limit of detection (LOD), limit of quantification (LOQ), and limit of linearity (LOL) were found to be 0.031, 0.1023, and 275 mg/ml, respectively. The proposed method was successfully applied in quantitative determination of 2′,4′-dihydroxy-6′-methoxy-3′,5′-dimethylchalcone in buds and leaves of C. operculatus collected from different places in Vietnam, which range from 13.24 to 18.61 and from 4.33 to 4.79 mg/g, respectively.
