Propagation of Malaleuca alternifolia Tree Introduced from Australia by Tissue Culture
In this study, an efficient protocol of Malaleuca alternifolia tree propagation using tissue culture technology was developed. Apical shoots explants (2-3 cm long) were collected from four-year old trees were used as initial materials for tissue culture. Explants were sterilized with HgCl2solution (0.1%) for 10 minutes and then with Javel solution (75%) for 15 minutes. Explants were cultured on LV medium containing BA (0.5 mg/l) for the highest shoot formation and multiplication. Regenerated shoots were rooted on LV medium supplemented with α-NAA (2 mg/l) giving the highest frequency (60.42%) after 4 weeks of culture. Whole plants were successfully transferred to soil pots and maintained in the green house before transferring to the field.
