Summary

Development of HPLC-PDA Method for Simultaneous Quantification of Caffeoylquinic Acid Derivatives

in the Fruits of Xanthium strumarium

A high-performance liquid chromatography (HPLC) method to simultaneously determine the content of chlorogenic acid, caffeic acid and 1,5-O-dicaffeoylquinic acid in the fruits of Xanthium strumarium was developed and validated. The samples were analyzed on a YMC-pack ODS-A column (150 x 4.6 mm, 5 µm) using mobile phase of acetonitrile - 0.1% aqueous formic acid in gradient condition, the flow rate of 0.8 mL/min, column temperature of 30oC, and detected wavelength of 327 nm. All calibration curves displayed a good linear relationship (r2 > 0.999). The validation results showed good accuracy, repeatability, linearity, and selectivity according to the ICH guideline. The validated method was then applied for the quantitative analysis of samples in Phu Tho province. The contents of marker compounds showed variable among collected samples as chlorogenic acid accounted for 0.17 - 0.25 %, caffeic acid 0.055 - 0.14%, and 1,5-O-dicaffeoylquinic acid were 0.14 - 0.24%.
