ĐỊNH LƯỢNG ROTUNDIN TRONG MỘT SỐ LOÀI THUỘC CHI STEPHANIA THU HÁI Ở VIỆT NAM BẰNG PHƯƠNG PHÁP HPLC-DAD

Hàm lượng rotundin trong bình vôi được xác định bằng phương pháp sắc ký lỏng hiệu năng cao kết hợp với detector DAD (HPLC-DAD), sử dụng cột sắc ký pha đảo Agilent XDB C₁₈, rửa giải bằng hệ gradient acetonitril (A) và 0,1% phosphoric acid trong nước (B), tốc độ dòng 1,0 mL/phút tại bước sóng 281 nm, khoảng nồng độ tuyến tính 0,5-50 µg/mL. Phương pháp định lượng được thẩm định cho thấy tính chọn lọc, đặc hiệu, chính xác phù hợp để định lượng rotundin trong dược liệu bình vôi. Kết quả định lượng một số mẫu bình vôi trồng ở Việt Nam cho thấy có sự khác biệt rất lớn về hàm lượng rotundin, dao động từ 0,25% đến 5,23% tính theo dược liệu khô kiệt. Trong đó, hai mẫu có hàm lượng rotundin cao nhất đều thuộc loài Stephania brachyandra (5,23% và 3,87%). Hai mẫu thuộc loài S. pierei không phát hiện được sự có mặt của rotundin. Các mẫu khác có hàm lượng rotundin dao động từ 0,25-1,65%.

Summary

Quantitative Determination of Rotundine in Stephania Species Collected in Vietnam Using HPLC/DAD Method

Rotundine content in Stephania species was quantitatively determined using an HPLC-DAD method, using an Agilent XDB C₁₈ reversed-phase column, a gradient condition of acetonitrile (A) and 0,1% phosphoric acid in water (B). The flow rate was 1.0 mL/min and the detection wavelengths were set at 281 nm, the standard curves were linear over the concentration range of 0.5-50 µg/mL. The method was then applied to determine the quantity of rotundine in Stephania species grown in different areas of Vietnam. The contents of rotundine in the indicated species were compared. The results revealed that the content of rotundine varied from 0.25% to 5.23% depends on the species of the analyzed samples. In which, two samples had the highest contents of rotundine, belongs to S. brachyandra species (5.23 ± 0.07 %) and (3.87 ± 0.08%). There was no rotundine in two samples of S. pierei species. In other species, the contents of rotundine varied from 0.25% to 1.65%.
