Summary

Extraction and Quantification of Hydroxychavicol in Betel’s Leaves (Piper betle L. Piperaceae) by HPLC - PDA

Hydroxychavicol (HC) is the major substance in betel’s leaves that can inhibit Candida spp. Betel’s extract was extracted by boiling with water at 1:5 (w/v) for 3 hours at a temperature of 1000C. The liquid extract was concentrated to 1/10 original volume, then purified by alcohol precipitation method at the ratio of the concentrated liquid and 96% of EtOH was 1:16 (v/v). The extract resisted Candida spp. at 2 mg/mL of MIC value. HC in the extract was quantified by RP - HPLC - PDA with SphereClone ODS (2) (4 x 250 mm, 5 μm) column. The isocratic mobile phase was ACN - 0.02% of H3PO4 (25:75) at a flow rate of at 1 mL min-1. The injected volume was 10 μL and HC was detected at 280 nm of wavelength. That analytical method showed linearity within the concentration of 1 - 10 ppm (R2 = 0.9996). The precision indicated the RSD of the percentage of HC in the extract was less than 3%. The percentage recovery of HC at 3 concentrations was in the range of 101.6 - 105.2%. The extraction and quantification were easy to apply for its standardization.
