HPLC ANALYSIS OF LOBETYOLIN IN RADIX CODONOPSIS JAVANICAE

Summary

HPLC Analysis of Lobetyolin in Radix Codonopsis javanicae

 

Codonopsis javanica roots have long been used as a popular remedy in Vietnam traditional medicine. Despite its popularity, the current monograph of Radix Codonopsis javanicae in Vietnamese Pharmacopoeia V (2017) only describes TLC qualitative analysis of lobetyolin, but no assay is mentioned. This study, therefore, aimed to develop and validate a reliable and effective quantitative HPLC-UV method for assaying lobetyolin, the officially accepted biomarker of Radix Codonopsis javanicae. The chromatographic separation was performed on Luna C18 column using gradient elution of water and acetonitrile, while the method was validated in terms of specificity, linearity (r2 > 0.999), precision [relative standard deviation (RSD) < 1.19%], and accuracy (98 - 102%). This validated method was subsequently applied to quantify the lobetyolin content in 2- and 3- year roots of Codonopsis javanica collected in Lam Dong province. The result showed that the HPLC-UV method was proven to be effective for the assay and the average content of lobetyolin in the 2- and 3-year Codonopsis roots were 0.066% and 0.206%, respectively. Thus, the established HPLC-UV method can be useful for assaying lobetyolin in the crude drug.
