HPTLC DENSITOMETRY METHOD FOR THE QUANTIFICATION OF NYSTOSE IN RADIX MORINDAE OFFICINALICOLLECTED FROM DIFFERENT REGIONS IN VIETNAM

HPTLC Densitometry Method for the Quantification of Nystose in Radix Morindae officinalis
Collected from Different Regions in Vietnam
In this study, a simple, selective, precise and stability indicating high performance thin layer chromatographic method for quantitative analysis of nystose in Radix Morindae officinalis (RMO) was developed and validated. The method employed TLC aluminium plates precoated with silica gel 60F-254 as the stationary phase. The solvent system consisted of ethyl acetate- propanol- water- glacial acetic acid (1: 1: 0.5: 0.5 v/v/v/v). Densitometric analysis of nystose was performed at 570 nm after derivatized with α-naphthol reagent and heated at 105oC for 3 min. The system was found to have compact spot for nystose [A1] with Rf value of 0.4 ± 0.02. For the proposed procedure, linearity (r2 = 0.996), limit of detection 0.04 µg/spot, limit of quantification 0.12 µg/spot; recovery (ranging from 96.92% - 97.30%) were found to be satisfactory. The method is rapid, precision, repeatability and accuracy for quantitative determination of nystose in RMO. Statistical analysis revealed that the content of nystose in RMO collected from different regions in Vietnam, ranging from 2.11 - 7.23%.
