Summary
Isolation and Establishment of Lobetyolin Reference Standard from Radix Codonopsis javanicae
Lobetyolin is a marker for the quality control of Radix Codonopsis javanicae. This herbal has long been a Vietnamese traditional medicine, which has been used for tonifying the middle Jiao, augmenting Qi, and tonifying the lung. However, the lobetyolin reference standard made in Vietnam has thus far been unavailable, which causes difficulties in the quality control practice of this valuable herb. In this study, we aimed at isolating lobetyolin from Radix Codonopsis javanicae by using reflux extraction method and different chromatography techniques including thin layer chromatography (TLC), column chromatography (CC) and preparative high performance liquid chromatography (prep-HPLC). As a result, lobetyolin (1.0 g) was isolated from 10.0 kg of Radix Codonopsis javanicae. The isolated compound was established as a reference standard based on ISO Guide 34 and 35. The adjust content of lobetyolin was determined to be 97.83%.
