QUANTIFICATION OF 2,7b,8b -TRIHYDROXYCALAMENENE-7-O-β-D-GLUCOPYRANOSIDE IN ABELMOSCHUS MOSCHATUS SUBSP. TUBEROSUS (SPAN.) BORSS. WAALK. BY HPLC-PDA

Summary

Quantification of 2,7b,8b -Trihydroxycalamenene-7-O-b-D-Glucopyranoside in Abelmoschus moschatus Subsp. tuberosus (Span.) Borss. Waalk. by HPLC-PDA

Abelmoschus moschatus subsp. tuberosus is a quite common medicinal plant in Vietnam but there are only a few of reports on its chemical components and bioactivities. In fact, there is no report on a HPLC method of quantifying any compound in this medicinal herb. In this study, a effective HPLC-PDA method of quantifying 2,7β,8β-trihydroxycalamenene-7-O-β-D-glucopyranoside in this species was developed and validated. The HPLC analysis was performed on a Lichrospher RP-18 column at 25oC, detection wavelength at 220nm, an injection volume of 20 µl and a flow rate of 0.9 ml/min with mobile phase of methanol and 0.1% formic acid (57/43, v/v). Specificity showed no interference from components of the mobile phase and of the extract of medicinal herbs. The method proved to be linear in the range 4.84 - 58.13 µg/ml. The limits of detection and quantification were 0.12 and 0.4 µg/ml, respectively. Repeatability and inter-day precision evaluated by relative standard deviation were below 3.0 %. Accuracy was accessed by the recovery test and achieved 98.97 - 101.23 % at 3 levels of added standard amounts. The content of 2,7β,8β-trihydroxycalamenene-7-O-β-D-glucopyranoside in Abelmoschus moschatus subsp. tuberosus was about 0.0213 %. This HPLC method may be used to control the quality of Abelmoschus moschatus subsp. tuberosus using 2,7β,8β-trihydroxycalamenene-7-O-β-D-glucopyranoside as a chemical marker.
