Quantification of Ursolic Acid in Oldenlandia corymbosa L. by HPLC-PDA

Oldenlandia corymbosa is a common medicinal plant in Vietnam but is not recorded in both Chinese and Vietnamese pharmacopoeias. In the world, reports of quantifying ursolic acid (UA) in Oldenlandia corymbosa by HPLC are also very limited. In this study, an effective HPLC-PDA method of quantifying UA in this species was developed and validated. The HPLC analysis was performed on a Lichrospher RP-18 column at 25oC, detection wavelength at 210nm, an injection volume of 10 µl and a flow rate of 0.4 ml/min with mobile phase of methanol and 0.05% phosphoric acid (91/9, v/v). Specificity showed no interference from components of the mobile phase and of the extract of medicinal herbs. The method proved to be linear in the range of 18.29 - 182.92 µg/ml. The limits of determination and quantification were 0.9 and 3.0 µg/ml, respectively. Intra-day precision evaluated by relative standard deviation was below 3.0 %. Accuracy was accessed by the recovery test and achieved 95.30 - 102.49%, an average 98.26%. The method was successfully employed to determine the contents of ursolic acid in four samples of Oldenlandia corymbosa collected.
