Summary

Quantitative Determination of Mimosine in the Medicinal Herb Mimosa pudica Mimosaceae by HPLC-PDA

An HPLC method was proposed for quantification of mimosine in the aerial parts of Mimosa pudica. The chromatography was established as: using the C18 (250 mm × 4.6 mm, 5 µm) column; the PDA detector was set at 275 nm; the mobile phase consisted of methanol and pH 2.0 buffer with ion-pairing agent sodium hexane-1-sulfonate, gradient elution; flow rate was 1 mL/min; the column temperature was set at 35°C; injection volume was 20 µL. The method was validated for the system suitability (RSD of peak area = 1.18%; RSD of tR = 0.03%); specificity; linearity ranging from 0.5 - 20.0 µg/mL (R2 = 1), precision (RSD < 3.0%) and accuracy (recovery: 92.43% - 98.11%). As for practical application, the tested samples showed the content of mimosine ranging from 64.5 µg/g to 363.1 µg/g, on dried basis.
