Bioassay-Guided Isolation of an Antioxidative and Hepatoprotective
Constituent from Orthosiphon aristatus
A preliminary screening on antioxidative activity of fractions from the medicinal plantOrthosiphon aristatus Blume showed that the ethyl acetate fraction (Et-F) possessed significant activity. Among 8 sub-fractions separated from the Et-F, sub-fraction 3 (Subf.3) exhibited the strongest activity against FRAP and DPPH in vitro assays, and ex vivohepatoprotective assay. Bioassay-guided fractionation and isolation on this subfraction resulted in the isolation of a pure compound, which was identified as rosmarinic acid by means of MS and NMR spectroscopic data. The isolated compound showed remarkable hepatoprotective activity on both ex vivo and in vivo assays in mice’s injured liver cells induced by carbon tetrachloride, in comparison with that of silymarin. These observations suggest that O. aristatus(as well as one of its principle compound rosmarinic acid) could be used as potential agent forprevention and treatment of oxidative liver damages beside its already known activities andusages.
