Determination of Some Active Components in Phyllanthus amarus Schum. et Thonn. by TLC - Scanning Method for Standardization
Thin layer chromatography- fingerprint (TLC-FP) analysis of lignan, flavonoid, phenolic compounds and quantitative determination of phyllanthin and lignan in the raw materials of Phyllanthus amarus Schum.et Thonn were performed. The solvent systems of n.hexan: ethylacetat (2: 1), ethylacetat: acetic acid: formic acid : water (10: 1: 1: 2) and the reagents of vanillin sulphuric acid, NP/ PEG and fast blue salts were applied for identification of lignan, flavonoids and phenolic compound respectively. For the quantitative analysis, HPTLC plate silicagel 60 Merck and the same solvent system for the identification of lignan were used. The derivative products of phyllanthin and lignan obtained by 10% sulphuric acid solution in ethanol and heat at 105oC for 20 minute were measure at l 610nm by scanning method. The calibration plot of phyllanthin is linear in the range of 0,1 – 0,8 mg ( r: 0,99943. The repeatability and the average recovery of the method is 2,26% (RSD) and 103,0%, respectively. The methods have contribuited to standardization research and quality control of the plant drug from Phyllanthus amarus Schum.et Thonn.
