Analysis of tanshinon IIA in mixture extract from Radix Salviae miltiorrhizae and Radix Astragali by Thin Layer Chromatography (TLC) and High Performance Liquid Chromatography (HPLC)
Tanshinon IIA is the main chemical components related to the bioactivities and pharmacological properties of Radix Salviae miltiorrhizae. This paper describes analysis of tanshinon IIA in mixture extract from Radix Salviae miltiorrhizae and Radix Astragali by TLC and HPLC.Chromatography was performed on a 4.6 x 250nm i.d., 5μm particle size, C18 column. The mobile phase was a linear gradient prepared from methanol : water (80:20) at a flow - rate of 1.5 ml/min, injection volume: 20 μl and UV detector: 270 nm. Good linearity (R2= 0.999) was observed over the concentration ranges investigated. Recovery was from 96.4 to 101.9 % and relative standard deviation were always < 3%. Tanshinon IIA in mixture extract from Radix Salviae miltiorrhizae and Radix Astragali was determined under the above analytical conditions, the percentage extraction of tanshinon IIA 0.559 % (w/w) in mixture extraction using ethanol concentration of 80% (v/v) is higher than other extraction solvents.
