Comparing Results of Ion-Exchange Chromatography Carried out on FPLC System with Manually Operated Separation Process

In this paper chromatographic separation of Vietnam cobra (Naja naja) venom ribonuclease (RNase) on CM-cellulose column (Æ0.9´11 cm) was carried out on FPLC system in the same conditions that were used in manually-operated column (Æ1.8´17 cm). It seemed that chomatographic separation profiles on these two methods are similar qualitatively: there are 2¸3 molecular forms of cobra venom RNase eluted from each of the columns. But careful analysis of the separation profiles showed that manners by which chomatographic processes were carried out influenced  on separation results considerably, namely revealed their 2 quantitative differences. Firstly, salt concentrations eluting enzym forms from FPLC-operated column are much lower than those for manually-operated one. Secondly, from FPLC-operated column were almost always eluted 3 RNase peaks, while manually-operated column often separated RNase from cobra venom into two molecular forms. In the more higher resolution chromatographic conditions FPLC-operated column could separate the enzym into 5 peaks.
