Summary

Study on Isolation and Standard Establishment of Sinensetin
from Extractum Herba Orthosiphonis spiralis

Orthosiphon spiralis, a well-known herbal medicine, is used for clearing Damp-Heat in the kidneys, bladder, promoting urination, and reducing kidney stones to treat high blood pressure and relieving pain in rheumatoid arthritis. Sinensetin is an active compound of O. spiralis, used for quality control of O. spiralis. The study aimed to isolate sinensetin from Extractum Herba Orthosiphonis spiralis using different chromatography techniques such as thin-layer chromatography, column chromatography, and preparative HPLC. The isolated sinensetin was demonstrated to meet the requirement of a reference standard based on ISO Guide 34 and 35. From 2.2 kg of Extractum Herba Orthosiphonis spiralis, 1.17 g of the isolated sinensetin was obtained with the sinensetin content of 99.83%.
