DEVELOPMENT OF AN ANALYSIS METHOD USING HPLC-UV FOR SIMULTANEOUS QUANTIFICATION OF ORCINOL GLYCOSIDE AND CURCULIGOSIDE IN RHIZOMA CURCULIGINIS

Development of an Analysis Method Using HPLC-UV for Simultaneous Quantification of Orcinol Glycoside and Curculigoside in Rhizoma Curculiginis
In this study, a simple and sensitive high performance liquid chromatographic (HPLC) method for simultaneous analysis of orcinol glucoside (OG) and curculigoside (CG) in Rhizoma Curculiginis was developed and fully validated. The separation was carried out on a column Ascentis C18 (250 mm × 4.6 mm; 5 µm) with a mobile phase consisting of acetonitrile - aqueous phosphoric acid 0.01% (21: 79, v/v) at a flow rate of 1 mL/min. The validation results showed good linearities (R2=1 for OG, R2=0.999 for CG); limits of detection and limits of quantification were 0.09 µg/mL and 0.2 µg/mL for OG, 1 µg/mL and 3.3 µg/mL for CG); recoveries ranged from 96.4 - 104.8% for OG and 95.1 - 101.3% for CG; repeatability, accuracy with relative standard deviations (RSDs) of less than 2.0%. This method is suitable for simultaneous quantitative analysis of OG and CG in Rhizoma Curculiginis. The obtained results provide useful information for the quality evaluation of Rhizoma Curculiginis in Vietnam.
