Quantitative Determination of Total Flavonoids in the Buds and Leaves of Cleistocalyx operculatus
by Colorimetric Method
Previous studies indicated that flavonoids are major compounds in the buds and leaves of Cleistocalyx operculatus, a medicinal plant has been used in Vietnamese and Chinese traditional medicine for long time. This study deals with the quantitative determination of flavonoids in these materials collected in the North of Vietnam. Total flavonoids were colorimetrically determined at the wavelength 510 nm based on the reaction with AlCl3. The method showed good linearity in the range 45-450 mg/l (R2 = 0.9993), as well as good recovery and accuracy. The limit of detection (LOD), limit of quantification (LOQ), and limit of linearity (LOL) were found to be 1.05; 3.51, and 1000 mg/l, respectively. The proposed method was successfully applied in quantitative determination total flavonoids in buds and leaves of C. operculatus collected from different places in Vietnam, which range from 3.48 to 4.21% and from 2.07 to 3.02 %, respectively.
