Summary

Method Development for Quantitation of Quercitrin in Houttuynia cordata Thunb. by HPLC

Houttuynia cordata Thunb. (Saururaceae) have been traditionally used as medicinal foods in South East Asia countries and Vietnam as well. Many studies have shown that flavonoids in H. cordata exhibited a wide range of biological effects including anti-inflammatory, anti-allergic, and anti-cancer activities because of their antioxidant properties. A rapid and sensitive HPLC method with UV detection was developed for the analysis and quantification of quercitrin (quercetin-3-O-a-L-rhamnopyranosid) - a major bioactive flavonoid in Houttuynia herb. Assay was conducted using a Symmetry C18 column (5 µm, 4.6 x 150 mm, Waters) under isocratic elution with 0.1% formic acid/water - acetonitril (80:20) at flow rate of 1 ml/min and detected using photodiode array at 265 nm. The method has a dynamic linearity in the range 25 - 300 µg/ml. The accuracy was in the range of 92 - 106% and the precision was 3.3%. The lower limit of detection (LLOD) was 25 µg/ml with the regression coefficient r2 = 0.996. The yield of quercitrin in aerial part ofHouttuynia cordata collected in different regions varies from 0.25 - 1.2%. The highest in sample from Quang Nam province and the lowest in sample from Ho Chi Minh city. HPLC fingerprint analysis also indicates that the flavonoid compositions of H. cordata in different areas of Vietnam had the same chromatographic pattern but altered by the amount of some main constituents.
