Study on Antioxidant Effect and Cytotoxic Activity of Trametes versicolor
The purpose of this study was to evaluate the antioxidant and cytotoxic activities of fractionated extracts (crude polysaccharide extract, MeOH extract, diethyl ether, n-butanol fraction and aqueous fraction) derived from Trametes versicolor. MeOH extract and n-butanol fraction showed the free radical scavenging effect on DPPH test and the anti-lipid peroxidation effect on MDA test. Crude polysaccharide extract showed only DPPH scavenging activity. Treatment of both cell lines with varying concentrations of aqueous fraction in culture resulted in a significant inhibition of tumor proliferation. Crude polysaccharide inhibited only the proliferation of tumor cell line A549. Furthermore, measurements of cell viability clearly showed that aqueous fraction was cytotoxic. Crude polysaccharide at the oral dose of 100 mg/kg for 8 days alleviated the increase of hepatic MDA content and restored the decrease of hepatic glutathione content caused by cyclophosphamide. These results demonstrated that Trametes versicolor was cytotoxic to tumor cell lines and produced an antioxidant effect on cyclophosphamide-induced liver damage
