\n\tIdentification of Drug- Metabolizing Enzymes on Rabbit Liver
\n\tSilymarin- induced Microsomal P450 by Application of two-Dimensional nano Liquid Chromatography Coupled online with Tandem Mass Spectrometry
\n\tSilymarin was first isolated from an extract of milk thistle fruit some three decades ago. Silymarin has been used to treat liver diseases for hundreds of year. Although regarded as safe, many of the extract constituents remain thus far untested for their possible effects on liver biotransformation enzymes, especially P450. In this report, the results showed that SDS-PAGE of the purified P450 solution showed four major bands from 49.0 kDa to 55.0 kDa. In the mascot, two enzymes namely expoxide hydrolase (52kDa) and carboxyl esterase (62kDa) by application of two-dimensional nano liquid chromatography (2DLC) coupled online with tandem mass spectrometry (MS/MS) were studied. Expoxide hydrolase and carboxyl esterase are the enzymes involving major phase II detoxification activities.
