SIMULTANEOUS QUANTITATIVE DETERMINATION OF LIMONIN, EVODIAMINE AND RUTAECARPINE IN EVODIAE FRUCTUS BY HPLC/DAD

Summary
Simultaneous Quantitative Determination of Limonin, Evodiamine and Rutaecarpine
in Evodiae fructus by HPLC/DAD
A simple HPLC method was developed for simultaneous determination of three major components, limonin, evodiamine and rutaecarpine in Evodiae fructus. The method was carried out by using a Gemini RP C18 column with a gradient solvent system of acetonitrile – methanol – water and a PDA detector (210 nm). The contents of the major components in Evodiae fructus extract could easily be determined within 30 minutes. All calibration curves showed good linear regression (r > 0.999) within the concentration ranges of 5 – 100μg/ml for evodiamine, 5-200 μg/ml for limonin and rutaecarpine. The RSDs of intra-day and inter-day were 4.81 – 6.07 %; 6.01-6.79 and 4.81 – 6.13 % for limonin, evodiamine and rutaecarpine, respectively. The LOD was 0.0625 μg/ml for 3 components and the LOQ were 0.125 and 0.2 μg/ml, for limonin, rutaecarpine and evodiamine, respectively. The recovery was 102.73% for limonin, 101.58 for evodiamine and 103.34% for rutaecarpine. This study provides a qualitative and quantitative method for analysis of limonin, evodiamine and rutaecarpine in Evodiae fructus. The developed method has been successfully applied to simultaneously evaluate the quality of different Evodiae Fructus in the market.
