Tóm tắt

Nghiên cứu này đánh giá tác dụng cải thiện trí nhớ của cao cồn rễ đinh lăng (Polyscias fruticosa (L.) Harms) trên mô hình thiếu máu não cục bộ tạm thời bằng mô hình thắt tạm thời 2 động mạch cảnh chung (T2VO), đồng thời bước đầu đánh giá cơ chế tác dụng của cao cồn rễ đinh lăng. Chuột nhắt trắng uống cao chiết cồn rễ đinh lăng với liều tương đương 300 mg dược liệu/kg trước khi tiến hành phẫu thuật gây suy giảm trí nhớ 1 tuần và trong suốt thời gian làm thí nghiệm. Tacrin (liều 2,5 mg/kg; i.p) được sử dụng làm đối chứng dương. Kết quả cho thấy cao chiết cồn rễ đinh lăng có tác dụng cải thiện suy giảm trí nhớ trên mô hình T2VO, được đánh giá bằng thử nghiệm nhận diện đồ vật và thử nghiệm mê lộ chữ Y cải tiến. Mức độ biểu hiện của một số gen liên quan bao gồm gen ChAT, muscarinic M3, M5 (M3, M5) cũng được đánh giá trong nghiên cứu này. Kết quả cho thấy cao cồn rễ đinh lăng (liều tương đương 300 mg dl/kg) có tác dụng tăng biểu hiện gen ChAT và muscarinic M5 so với nhóm chứng bệnh lý (T2VO). Nghiên cứu của chúng tôi đã chứng minh cao chiết cồn rễ đinh lăng với liều tương đương 300 mg dl/kg có tác dụng cải thiện suy giảm trí nhớ thông qua cơ chế tăng cường biểu hiện gen ChAT và muscarinic M5.

Summary

Study on the Anti-dementia Effect of Ethanol Root Extract from Polyscias fruticosa (L.) Harms and its Mechanisms

This study evaluated the anti-dementia effects of Polyscias fruticosa (L.) Harms ethanol root extract, using transient 2 vessels occlusion (T2VO)-induced cognitive deficits in mice, and its mechanisms underlying the action. T2VO mice were treated daily with ethanol root extract of Polyscias fruticosa (300 mg dried root/kg body weight, p.o) 1 week before and continuously a day after surgery. Administration of the reference drug tacrine (2.5 mg/kg/day, i.p.) to T2VO mice was started a day after T2VO. Cognitive performances of the animals was analyzed using the novel object recognition test, modified Y maze test, to elucidate non-spatial short term memory and spatial working memory, respectively. After completing the behavioral experiments, the expression level of cholinergic marker genes, including choline acetyltransferase (ChAT), muscarinic M3 and M5. T2VO caused impairment of non-spatial short term memory and spatial working memory. T2VO -induced memory deficits were reversed by Polyscias fruticosa extract and tacrine. Polyscias fruticosa extract attenuated T2VO-induced down-regulation of choline acetyltransferase and muscarinic M5 genes expression without affecting muscarinic M3 gene. These finding demonstrate that Polyscias fruticosa extract improves memory deficits caused by T2VO and that the effect of Polyscias fruticosa extract may be mediated by enhancement gene expression of central cholinergic systems, such as ChAT and muscarinic M5.
