\n\tThe increasing prevalence of dental caries is making it more major problem of world health. Dental caries is the direct result of acid production by cariogenic oral bacteria. New and better antimicrobial agents active against cariogenic bacteria are badly needed, especially natural agents derived directly from plants. In this study, a-mangostin, a xanthone was isolated from n-hexane extracts of the tropical plant Garcinia mangostana L. grown in Vietnam by repeated silica gel chromatography and it was found to be a potent inhibitor of acid production by Streptococcus mutans UA159S. sobrinus 6715 and S. sanguis NCTC 10904, active against membrane enzymes responsible for acid production and tolerance, including the F(H+)-ATPase and phosphoenolpyruvate - sugar phosphotransferase system. Glycolysis by intact cells in suspensions or biofilms was inhibited by a-mangostin at concentrations of about 12 and 120 mM, respectivelyThe overall conclusion is that a-mangostin is an inhibitor of mutans Streptococci and could be useful as an anti-caries agent.
