Summary
In vitro Conservation of Dendrobium nobile Lindl. and Anoectochilus setaceus Blume. by Slow Growth Culture
The aim of this work was to develop protocols for slow growth conservation of Dendrobium nobile Lindl. and Anoectochilus setaceus Blume. Shoots of D. nobile were maintained at 250C in a 1/4 MS culture medium containing 5 g/l sucrose and that of A. setaceus in 1/2 MS with 10 mg/l sucrose. After a period of six months without any subculture, shoots of both species were recovered on full strength MS medium containing 30 g/l sucrose plus 6 g/l agar supplemented with a) 1 g/l pepton, 10% coconut water, and 0.5 mg/l Kin for D.nobile and b) 30 % potato extract and 1.5 mg/l Kin for A. setaceus. Retrieved shoots were raised into plantlets and acclimatized in nursery with high survival rates.
